A third pool contained three L-segment amplicons targeting outbreak-specific SNPs to increase sensitivity for outbreak specimens. Random priming used 65°C for 5 min followed by 2 min snap cooling at 4°C. cDNA was synthesized with SuperScript IV (Invitrogen, Carlsbad, USA) at 23°C for 10 min, 54°C for 15 min and 80°C for 10 min. Tiled ANDV amplification followed a modified ARTIC protocol V.4 (https://dx.doi.org/10.17504/protocols.io.bp2l6n26rgqe/v4), using the Q5 High-Fidelity DNA Polymerase Kit (Integrated DNA Technologies, Coralville, USA) in 50 µl reactions: 98°C for 30 s, then 38 cycles of 98°C for 15 s and 65°C for 5 min. Libraries were prepared from 100 ng input with the Native Barcoding Kit 24 v14 (SQK-NBD114.24) and sequenced on an R10.4.1 flow cell (FLO-PRO114M). Reads were base called with Dorado v.5.2 at super high accuracy, requiring barcodes at both ends (all Oxford Nanopore Technologies, Oxford, UK). 0	0	0	0	0	2	0	0	0	0	0	6	386	206	127	0	0	0	0	0	0 "[{""fileId"":""FILE_00A04S1"",""name"":""PP_00843Q0.1.embl"",""url"":""https://pathoplexus.org/seq/PP_00843Q0.1/annotations/PP_00843Q0.1.embl""}]"
